FIELD: steroids, organic and analytical chemistry, medicine, biology. SUBSTANCE: invention relates to the synthesized 5β-[4,5-3H]-pregnane-3,20-dione and 5α- [4,5-3H]-pregnane-3,20-dione highly labeled with tritium that are used for determination of metabolites of progesterone in blood plasma. Tritium-labeled analogues of progesterone metabolites to be assayed are added to analyzing sample of blood plasma. Chloroform extract contains both analyzing two metabolites and other metabolites of progesterone and progesterone that interferes with assay. These interfering steroids are decomposed by contact of extract with hydrogen peroxide in an alkaline medium. Metabolites remained in extract after this treatment are converted to dansyl derivatives that ensures to determine nanogram amount of these steroids by HPLC method. Based on data of determination of the content of dansyl derivatives the concentrations of analyzing metabolites in blood plasma sample are calculated.
