FIELD: medicine; pharmacology.
SUBSTANCE: invention relates to medicine, namely, to experimental cardiac pharmacology, and can be used in a comprehensive assessment of the activity of pharmacological agents on an isolated rat heart. For this, method includes the registration of indices of contractility under conditions of imposing a high rhythm of contractions and hypercalcium load on the heart. At the same time, the parameters of the contractile function of the left ventricle are recorded, then a 20-minute perfusion of the heart with a solution containing 5 mmol/l of Ca2+ is performed to determine the dynamics of the diastolic stress. In the future, the reduction frequency is set to 480 bpm for 15 seconds using an electric stimulator. Then the coefficient STTI (area of defect diastole) is calculated in conventional units by folding trapezium squares under the curve of elevation of the end-diastolic pressure and evaluating the contractile function of the heart, taking into account the physiological values of STTI.
EFFECT: method provides information and increase in the reliability of evaluation indicators of the degree of cardioprotective or cardiotoxic activity pharmacological means using the coefficient STTI.
