FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology, specifically to a serum-free nutrient medium, and includes a main component composition of Eagle MEM nutrient medium, and additionally contains microelements (cadmium, cobalt, zinc, nickel, selenium, molybdenum) salts in concentration of 0.00001–4.0 g/l, sodium pyruvate in concentration of 0.04–4.0 g/l, vitamins B12 and E in concentration of 0.0001–0.1 g/l, oleic acid in concentration of 0.000005–0.5 g/l, cysteine, glutathione, proline, alanine, asparagine, aspartic acid in concentration 0.001–1.5 g/l, (total 45 components), recombinant human insulin in concentration 0.000025–0.025 g/l, as well as increased concentration of leucine, phenylalanine, tryptophan, lysine to 0.035–0.072 g/l. Serum-free nutrient medium when culturing MDCK cells provides cell growth, cells have a morphology typical for the culture, a monolayer is formed on 2–3th day of growth, preserving high proliferative activity (cell culture cell proliferation index is 3.2). Production of measles virus in the Vero cell culture and influenza viruses in the MDCK cell culture in the declared serum-free medium is comparable to production of these viruses in the serum-free SFM4MegaVir medium.

EFFECT: invention enables to create a more standardized serum-free nutrient medium for culturing MDCK or Vero cells or vaccine strains of measles or influenza viruses in order to create vaccines.
