FIELD: nuclear medicine, oncology, pharmaceutical chemistry and pharmacology.
SUBSTANCE: invention relates to a method of producing a radiochemical compound based on iodine-123-labeled recombinant protein molecules with ankyrin repeats (DARPin) for radionuclide diagnosis of malignant tumours with overexpression of HER2/neu. According to the method, 2–20 mcg of sodium iodide and a 123I solution with an activity of 280–1,200 MBq are added to a vial containing 500–4,000 mcg of DARPin G3 protein, then the pH is adjusted to 5.5–7.5 with a solution of 0.1 M hydrochloric acid and incubated for 3–7 minutes. After this, 40–120 mcg of chloramine-T are added, mixed and incubated at room temperature for 60–240 s. Next, 80–240 mcg of sodium metabisulfite are added and purified using a Sephadex G25 gel filtration column. A sterile 0.9% sodium chloride solution is added to the purified solution and filtered through a sterilizing filter.
EFFECT: radiochemical yield of more than 95% is provided.
