FIELD: generic engineering. SUBSTANCE: invention relates to the preparing new plasmids containing synthetic DNA nucleotide sequence which can be used for expression of protein rseu-PA at preliminary stage (i. e. nonglycosylated part of a single-chain prourokinase) in enterobacteria, in part, in E. coli. Operon of these plasmids has the operated synthetic promoter, sequence Shine-Dalgarno, start codon, synthetic structural gene which is located at 5'--3'-direction from structural gene and at least one terminator. Protein expressed by plasmids is subjected for the reverse reaction up to preparing the plasminogen activator rseu-PA exhibiting therapeutic activity. This activator can be split for large-scale preparing double-chain nonglycosylated urokinase (rseu-PA). EFFECT: improved method of plasmid preparing. 1 tbl, 27 dwg
