FIELD: biotechnology, genetic engineering. SUBSTANCE: DNA fragment encoding gene of tRNA Cis-(AAA) of the identified nucleotide sequence is obtained by synthesis of oligomers 5'-3' and 3'-5' using Hind III/Pst I and oligomers 5'-3' and 3'-5' using Pst I/Bam HI. Oligomers are renaturated and formed a mixture for ligation of renaturated oligomers and plasmid pUC 18 cleaved by Hind III/Bam HI. Mixture is incubated and used for transformation of Escherichia coli strain. Strain is cultured on medium with ampicillin and X-gal. Transformed strains with plasmids containing DNA fragment are selected and their base sequences are determined. Invention provides synthesis of growing peptides on ribosomes of Escherichia coli using the obtained synthetic tRNA. EFFECT: improved method of preparing. 2 cl, 9 dwg, 4 tbl
