FIELD: biochemistry, biotechnology. SUBSTANCE: method involves incubation of ribosome samples with plasmid DNA encoding the required protein in cell-free protein system. Incubation is carried out in medium for coupled transcription/translation with aminoacyl-tRNA carrying the fluorescent label. Then obtained protein is purified, its amount is measured and fluorescent intensity and biological activity are determined. Incorporation of fluorescent label at N-terminal part of synthesized protein provides the reliable detection of synthesized proteins that retain their full enzymatic activity.
