METHOD OF CONTROL OF BIOGEL CYTOTOXICITY USING HUMAN TRANSPLANTABLE LEUKOCYTE STRAIN L-41-KL/84 Russian patent published in 2000 - IPC

FIELD: medicine, experimental virology, plastic surgery. SUBSTANCE: invention relates to method of evaluation of quality of polyacrylamide gels. Control of biogel cytotoxicity is carried out using the strain L-41-KL/84 of human transplantable leukocytes. Gel is added to medium of test-culture growth at the dose of cells 60 000 cells/ml and cultured for 72-96 h followed by assay of cytophysiological and cytomorphological stability of culture in dynamics. Morphological study is carried out by conventional procedure. Indices of attachment efficiency, proliferative and mitotic activity of cells, mitosis pathology and morphology of cells in culture are determined. Cytotoxicity of biogels is estimated by change of these indices as compared with control data. The same test-culture is used as control but without gel addition. Invention ensures to differentiate biogel preparations by their cytotoxicity that excludes the possibility of using cytotoxic and adulterated materials and prevents deleterious results of effect on body. Objectivity and standard of control of preparation quality provides the use of standard test-culture of human transplantable leukocytes of the strain L-41-KL/84 for evaluation of cytotoxicity being this strain shows stability of cultural and morphological traits. Morphological analysis ensures to evaluate the trend of cytotoxic effect of analyzed gel in determination of changes of mitotic activity, morphology, in part, the state of metaphase nuclei in cellular culture.
