FIELD: medicine, microbiology. SUBSTANCE: invention relates to preparing diagnosticum by culturing diphtheria microorganisms on the solid nutrient medium followed by preparing a bacterial suspension with density 1000 OU/ml, its heating at +56-58 C for 40 min, treatment with formalin and merthiolate, washing out with 0.15 M sodium chloride solution, disintegration, isolation from suspension a diphtheria antigen by dialysis through a membrane with pores 40-60 nm, dilution of an antigen to concentration 250-400 mcg/ml by protein and its combination in volume 1 ml with 3 ml of 50% formalinized sheep erythrocytes washed out with 0.15 M sodium chloride solution, 5 ml of 20% solution of polyethylene glycol with molecular mass 6000 kDa and 4 drops of formalin. Then the prepared suspension is kept at temperature +22-24 C for 2 h and at +4-8 C for 24 h. Invention can be used for diagnosis of diphtheria.
