SET OF RECOMBINANT PLASMID DNA PYAI 11-19, PYAI 2-45, PYS 37 AND PYAI 7-29 FOR DETERMINATION OF ORIGIN OF HUMAN ACCESSORY OR MARKER (MINI-) CHROMOSOMES Russian patent published in 2003 - IPC

Abstract RU 2200761 C1

FIELD: genetic engineering, medicinal genetics. SUBSTANCE: set of recombinant plasmid DNA consists of 4 plasmid DNA. pYAI 11-19 is genome fragment of human DNA of size 1500 nucleotide pairs and involves 4 internal unique restriction site Eco RI. DNA fragment is limited by Pst I-sites from different ends of genome fragment. Under "mild" conditions this probe DNA is hybridized with centromere regions of chromosomes 1, 3, 5, 6, 7, 10, 12, 16, 19 or under "rigid" conditions with centromere region of chromosome 10 only. pYAI 2-45 is genome fragment of human DNA of size 1100 nucleotide pairs, involves 3 internal unique restriction site Eco RI and limited by Pst I-sites from different ends of genome fragment. Under "mild" conditions this probe DNA is hybridized with centromere regions of chromosomes 2, 4, 8, 9, 18, 20 but under "rigid" conditions with centromere region of chromosome 4 only. pYS 37 is genome fragment of human DNA of size 680 nucleotide pairs, involves 2 internal unique restriction sites Eco RI and limited by Pst I-sites from different ends of genome fragment. Under "mild" conditions this probe DNA is hybridized with centromere regions of chromosomes 13, 14, 15, 21, 22, Y but under "rigid" conditions with centromere regions of chromosomes 13 and 21 only. pYAI 7-29 is genome fragment of human DNA of size 2700 nucleotide pairs, involves 7 internal unique restriction sites Eco RI and limited by Pst I-sites from different ends of genome fragment. Under "mild" conditions this probe DNA is hybridized with centromere regions of chromosomes 1, 11, 17, X but under "rigid" conditions with centromere region of chromosome 17 only. Invention ensures to determine origin of human accessory or marker (mini-) chromosomes in practice of medicinal genetics, namely, in prenatal diagnosis, clinical cytogenetics and oncocytogenetics. EFFECT: improved method of determination. 3 ex
