FIELD: medicine, diagnostics. SUBSTANCE: human venous blood should be centrifuged at double density gradient of phycol- verografin 1.118 g/ml and 1.076 g/ml. One should perform hypotonic lysis of erythrocytic admixture, granulocytes are worked up to 5•106 cells/ml. concentration. Then it is necessary to determine the level of spontaneous and induced activity, moreover, as inductors one may apply opsonized and nonopsonized zymosan. At detecting the level of spontaneous activity one should apply 50 mcl 0.4%-NBT solution, 50 mcl incubation medium and 100 mcl cellular suspension into the hole; at determining the level of induced activity - 50 mcl NBT solution, 50 mcl stimulant's suspension and 100mcl cellular suspension; the plotting board with samples is incubated at 37 C, centrifuged, residue is obtained, ethanol is added to be recentrifuged, supernatant is repeatedly obtained, 0.85%- NaCl is added to be centrifuged again, then every sample is supplemented with dimexide to be incubated at 58-63 C, KOH solution is added, results of reaction are registered with photometer by extinction difference at 630 and 490 nm wave length.
