METHOD OF TESTING METABOLIC ACTIVITY OF BLOOD PHAGOCYTES BY NBT-TEST METHOD Russian patent published in 2013 - IPC G01N33/49

FIELD: medicine.

SUBSTANCE: sampling of 0.1 ml of blood from finger is performed, 0.9 ml of medium 199 is added to it, after which 0.1 ml of 0.1% of nitro blue tetrazolium and 0.1 latex suspension with particle size 1.5 mcm are added and incubation for 40 minutes at 37C is carried out. In order to obtain leukoconcentrate obtained mixture is additionally centrifuged, supernatant liquid is discharged, sediment is covered with 2 ml of fixer, which consists of 3 parts of ethanol and 1 part of icy acetic acid for 10 minutes. After that cell suspension is centrifuged, fixer is sucked out. Cells, resusended in fixer drop, are transferred onto microscope slide, obtained smear is dried, after which it is stained with solution of neutral red dye for 20 minutes. Then smear is microscoped under immersion with 90×10, magnification, results of tests are evaluated by calculation of percent of formazan-positive cells, calculation of cytochemical index and index of NBT-test stimulation.

EFFECT: application of claimed method makes it possible to obtain sufficient necessary for calculation concentration of phagocytes, which contributes to increase of test accuracy.
