FIELD: medicine.
SUBSTANCE: method involves using iron oxide Fe2O3 with immobilized specific antibodies as magnetic immunosorbent. 1 ml of corresponding antigen suspension is added to 0.2 ml of 10% iron oxide Fe2O3 suspension with immobilized specific antibodies. The mixture is incubated during 30-60 min. at 22±4°C. Then washing is carried out with 0.1 M phosphate salt buffer containing 0.5% twin-20 by means of permanent magnet. 200 mcl of peroxidase conjugate of working dilution is added and incubated at 22±4°C during 15 min. The washing is repeated. 200 mcl of chromogenic mixture is added and flask content is transferred to microplates in 1-2 min. The reaction is stopped with 50 mcl of sulfuric acid solution of 2 M. The results are recorded with spectrophotometer.
EFFECT: accelerated examination process; prolonged immobilized solid phase service life; high sensitivity and specificity.
