METHOD FOR EXTRACTION AND DIFFERENTIATION OF PHAGE VIBRIO MIMICUS Russian patent published in 2009 - IPC C12N1/00 C12Q1/02 

FIELD: biotechnologies.

SUBSTANCE: as indicator strain they use Vibrio cholerae eltor KM 199. Its three-hour broth culture in amount of 0.2 ml is introduced into mixture of incubated, filtered lysogenic microorganism: 1 ml of the latter is joined with 5 ml of 0.7% Martin's agar. Then suspension is seeded onto agar plate of 1.5% Martin's agar and cultivated at 37°C for 20-24 hours. By lytic response conclusion is made on availability of phage Vibrio mimicus F-19. At the same time its differentiation from cholera phage FB 0139 of serogroup is carried out with the help of strain Vibrio cholerae 1322-69-037 of serovar. They prepare 0.2 ml of four-hour broth culture of the latter, 5 ml of 0.7% agar is added and poured onto agar plate into Petri dish. After congelation, on one path they apply phage F-19, and on the second one - cholera phage FB. Then they are placed in thermostat and grown for 24-48 hours at temperature of 37°C. By registration of lysis on one of paths they decide on availability of phage Vibrio mimicus on it.

EFFECT: method is suggested for extraction and differentiation of phage vibrio mimicus.
