FIELD: medicine.
SUBSTANCE: there are involved preparing a base AI medium containing micro- and macroelements, iron sources, organic substances in the preset quantities of the ingredients in the appropriate MSG-modification of the medium composition. The prepared medium is added with explants of Siberian larch seed embryos selected at the stage of ripening of seed lobes and resistant to Dasyneura laricis. At the stages of somatic embryogenesis, the method provides additional inclusion of the following ingredients into the base AI medium: at the stage of initiation - mesoinosit - 1 g/l, 2,4D - 2 mg/l, 6-benzylaminopurine - 1 mg/ml, saccharose - 30 g/l, agar - 7 g/l; at the stage of proliferation - mesoinosit - 1 g/l, 2,4D - 2 mg/l, 6-benzylaminopurine - 0.5 mg/ml, saccharose - 20 g/l, Gellan gum - 4 g/l, casein hydrolysate decreased to 500 mg/l; at the stage of pre-ripening - mesoinosit - 1 g/l, saccharose - 30 g/l; at the stage of ripening - mesoinosit - 1 g/l; abscisic acid - 32 mg/l; indole butiric acid - 0.2 mg/l, saccharose - 40 g/l, Gellan gum - 4 g/l; at the stage of germination - Gellan gum - 4 g/l with the concentration of micro- and macroelements to be decreased twice and organic nitrogen and vitamin sources to be excluded.
EFFECT: there have been grown somatic plants regenerating Siberian larch, resistant to Dasyneura laricis and used to create healthy larch forests characterised by fast growth and high yield.
