FIELD: biotechnology.
SUBSTANCE: invention is a method for morphogenesis stimulation in a barley tissue culture, comprising callus obtaining on a dense Murasige-Skoog medium containing 2 mg/l of dichlorophenoxyacetic acid; passage for proliferation to the medium with 1 mg/l of dichlorophenoxyacetic acid, and three weeks later - for morphogenesis induction - passaging to the medium with 1 mg/l of kinetin, 0.5 mg/l of indolylacetic acid, 0.1 mg/l of gibberellic acid. One medium (for callus proliferation or morphogenesis induction) is prepared not dense, but semi-liquid (4 g/l of agar) with addition of 5 vol % of perfluorodecalin; callus cultivation in a semi-fluid medium at constant shaking (125 rpm) for 2-3 weeks prior to callus appearance and subsequent regeneration of plants on a dense medium.
EFFECT: invention allows to increase the yield of barley regeneration plants by stimulating the laying of meristematic zones in callus tissue and increasing the morphogenetic potential in callus lines with reduced regenerative capacity.
