FIELD: medicine.
SUBSTANCE: preparation of a single-cell suspension from tumour samples fixed in 4% formaldehyde solution, staining with a specific DRAQ7 DNA dye, flow cytometer analysis, construction of histograms of cell distribution by intensity of DNA staining in the WinMDI 2.9 program, placement of markers separating the peak corresponding to diploid cells in G0/G1 phases of the cell cycle from aneuploid cells and cells in M, S and G2 phases of the cycle, as well as the total number of cells, calculation of the integral cell index by the formula: I=M2/M1×100%, where I is the integral index of aneuploidy and proliferative activity of cells, M1 is the total number of tumour cells, M2 is the number of cells in the M, S and G2 phases of the cycle, regardless of ploidy, the integral index exceeding 30% indicates an unfavourable outcome and tumor resistance to chemotherapy, the total index value less than 30% indicates a favourable prognosis and sensitivity to chemotherapy.
EFFECT: assessment of the prediction of the disease course and tumour sensitivity to chemotherapy.
