SUBSTANCE: invention relates to biotechnology. Method for producing carboxypeptidase B from pig pancreas involves homogenization of animal feed, autolysis, double-volume extraction of a buffer solution containing 0.01 M KH2PO4 0.001 M ZnCl2 at a temperature of 65 °C to obtain an extract. Ammonium sulfate is precipitated in a predetermined ratio and centrifuged. Precipitate is dissolved in a 0.05 M Tris-HCl buffer containing 0.001 M ZnCl2, filtered through a glass fiber and cardboard filters at the specified temperature parameters. Filtrate is diluted in 0.005 M Tris-HCl buffer containing 0.001 M ZnCl2, followed by ultrafiltration in a tangential flow on a membrane cassette module of polyethersulfone with a cut-off limit of 20 kDa and double ion-exchange chromatography on a column with UNO Shhere. Balanced with 0.005 M Tris-HCl buffer containing 0.01 NaCl and on a Macro Prep Q column equilibrated with the same buffer.
EFFECT: invention allows to increase the yield of the enzyme and its activity.
