ENZYMATIC PERACID GENERATION FOR USE IN ORAL CARE PRODUCTS Russian patent published in 2018 - IPC A61K8/22 A61K8/37 A61K8/38 A61K8/60 A61K8/66 A61Q11/00

Abstract RU 2660373 C2

FIELD: hygiene.

SUBSTANCE: group of inventions relates to the field of personal care products intended for oral treatment with a peracid agent, a method for preparing such an agent and a perhydrolytic enzyme in the form of a fusion protein for preparation thereof. Peracid is obtained from a substrate using an enzyme catalyst in the form of a fusion protein having perhydrolytic activity in the presence of a source of peroxygen. Said fusion protein comprises the following general structure: PAH-[L]y-OCBD or OCBD-[L]y-PAH, where PAH is an enzyme having perhydrolytic activity, and is a carbohydrate esterase having a signature motif CE-7 that aligns with the reference sequence SEQ ID NO: 2 and contains: 1) the RGQ motif at positions corresponding to positions 118-120 of SEQ ID NO: 2; 2) the GXSQG motif at positions corresponding to positions 179-183 of SEQ ID NO: 2, and 3) the HE motif in positions corresponding to positions 298-299 of SEQ ID NO: 2; OCBD is a peptide component having an affinity for the surface of the oral cavity, and is an antibody from a single-stranded variable fragment (scFv) or a single-stranded polypeptide without immunoglobulin folding; L is an optional peptide linker with a length in the range of 1 to 100 amino acids; and y is 0 or 1. Oral care product comprises said fusion protein, at least one carboxylic acid ester substrate, a peroxygen source and an oral acceptable carrier medium.

EFFECT: peracid beneficial agent can be used for clarification, bleaching, decontamination, discolouration, deodorisation, reduction and/or removal of biofilm from the surface of the oral cavity, while the use of the above-mentioned fusion protein provides for "targeting" perhydrolytic activity to the surface of the oral cavity.
