FIELD: toxicology.
SUBSTANCE: invention relates to toxicology, namely to methods of determining N-(benzimidazolyl-2)-O-methylcarbamate in biological material. For this purpose, biological tissue is milled, extracted three times for 15 minutes with a mixture of solvents ethyl acetate-dichloroethane-formic acid at ratio of 5:5:1 by volume. Extracts are combined, filtered and acidified with 0.1 n. solution of HCl. Then, the acidic extract is washed with ethyl acetate, the aqueous layer is alkalised with 10 % NaOH solution till pH 8–9 and repeatedly extracted with ethyl acetate. Extract is separated, dehydrated and dried by evaporation of solvent. Residue is dissolved in formic acid and chromatographed in a macrocolumn with Silasorb C-8 15μ sorbent, mobile phase: acetonitrile-water in ratio of 4:6 by volume. Fractions containing the analysed substance are combined, treated with ammonia solution and evaporated to dry residue. Residue is dissolved in a mixture of dioxane-propanol-2 in ratio of 5:1 by volume. Determination is carried out by HPLC with UV detector on column with sorbent Silasorb 600, 64×2 mm, mobile phase: hexane-dioxane-propanol-2 with ratio of 15:5:1 by volume.
EFFECT: invention provides a quantitative determination of a pesticide.
