FIELD: radiation biology.
SUBSTANCE: invention relates to radiation biology, namely to indication of radiotoxins in irradiated food and fodder products for assessment of their biological safety. That is ensured by preparing the analyzed material and homogenising the samples in the presence of an extractant. Concentration is carried out by evaporation on a vacuum evaporator of ethanol extract precipitate subjected to physiological saline dilution of serological analysis. Antibody bentonite diagnosticum is prepared on the basis of antiradiotoxic serum to produce bentonite nanoparticles by dispersing the powder in an ultrasonic unit. Obtained complex is labeled with methylene blue and bentonite flocculation reactions in crosses are visually evaluated. Serological analysis is carried out in a bentonite flocculation reaction (BFR) with an antiradiotoxic bentonite diagnosticum (ATBD). Carrier of specific antibodies is represented by bentonite nanoparticles sensitized by antiradotoxic serum with size of 75–85 nm, for sensitization of which hyperimmune serums of rabbits are used. Flotation of sensitized bentonite nanoparticles in BFR is used to assess the presence of radiotoxins in the analyzed sample. Vegetable radiotoxin is used as an immunising agent-antigen. Concentration of radiotoxins is determined by maximum dilution of the analyzed material – 1:8 and lower, indicating ecological safety of food and fodder products.
EFFECT: invention increases sensitivity of the indirect hemagglutination reaction (IHAR) test system by 2 times and the BFR test system by 1,66 times compared to the prototype.
