PROTON-BINDING POLYMERS FOR ORAL ADMINISTRATION Russian patent published in 2020 - IPC A61K31/785 A61P3/12

Abstract RU 2728778 C2

FIELD: medicine; pharmaceuticals.

SUBSTANCE: group of inventions relates to the treatment of metabolic acidosis. Disclosed is use of a proton-binding cross-linked amine polymer in the manufacture of a medicament for treating metabolic acidosis, wherein the cross-linked amine polymer includes residues of 2-propen-1-ylamine or a salt thereof, 1,3-bis(allylamino)propane or its salt, and 1,2-dichloroethane, wherein the cross-linked amine polymer is obtained via radical polymerisation of 2-propen-1-ylamine or its salt and 1,3-bis(allylamino)propane or its salt, followed by cross-linking with 1,2-dichloroethane after polymerisation, and where the cross-linked amine polymer has (i) an equilibrium proton-binding capacity of at least 5 mmol/g and a chloride-ion storage capacity, at least 5 mmol/g in an aqueous buffer solution simulating gastric juice ("SGF") containing 35 mM NaCl and 63 mM HCl at pH 1.2 and 37 °C, (ii) equilibrium swelling ratio in deionized water of 2 or less, and (iii) a molar ratio of binding of chloride ions to phosphate ions of at least 1:1, respectively, in an aqueous inorganic buffer solution which imitates a medium in the small intestine ("SIB"), containing 36 mM NaCl, 20 mM NaH2PO4 and 50 mM 2-(N-morpholino)ethane sulphonic acid (MES), buffered to pH 5.5 and at 37 °C. Invention also discloses a pharmaceutical composition for treating metabolic acidosis and a method of treating metabolic acidosis.

EFFECT: group of inventions provides safe and effective treatment of metabolic acidosis without introducing harmful counterions.
