FIELD: biotechnology.

SUBSTANCE: invention relates to separation of factor VIII protein (FVIII) from von Willebrand factor protein (vWF) and production of modified blood plasma, and can be used in medicine for treating diseases, requiring coagulation activity reduction, or FVIII activity determination in the sample. Methods include bringing a first composition, e.g. blood plasma containing FVIII and vWF, into contact with VIIISelect affinity resin containing a FVIII-affinity ligand, and a matrix, separating affinity resin from the mixture to produce a modified first composition and a second composition containing an affinity resin and a FVIII and vWF complex. Amount of VWF in the first composition is higher than the amount of FVIII protein, and the ratio of the volume of the first composition to the volume of the resin is in range of 5:1 to 70:1.

EFFECT: invention enables to obtain a modified blood plasma in which the FVIII concentration is less than 4 % of the average plasma FVIII concentration in a healthy human donor.
