METHOD FOR PREPARATION OF MYCELIUM OF BASIDIOMYCETES FOR SOIL MYCORIZATION IN ARTIFICIAL CULTIVATION OF CONIFEROUS PLANTS Russian patent published in 2021 - IPC A01H1/00

FIELD: agricultural biotechnology.

SUBSTANCE: developed method is based on the use of mycelial cultures of basidiomycetes of three types of fungi: Tricholoma equestre, Lactarius rufus, Suillus bovinus, which are promising mycorrhizal agents. Mycelial cultures were obtained by growing on Petri dishes with wort-agar containing beer wort at a concentration of 8° according to Balling, in a thermostat (“Thermostat TS-200 SPU”, Russia) at a temperature of 25±1°C for 5 days. Grown mycelial cultures are washed off from the surface of the medium with 10 ml of beer wort at a concentration of 8° according to Balling and are subject to homogenization at a mode of 8000 rpm for 2 minutes. Suspensions of homogenized mycelium fragments of each fungus type are brought to the concentration 10 IU of the turbidity standard OSO 42-28-85-2014 with 0.9% sodium chloride solution. From the obtained suspension, for T. Equestre and L. Rufus fungi, the 5th dilution is prepared and for S. bovinus fungus, the 8th dilution of a suspension of mycelium fragments is prepared, and the dilutions in a volume of 7.5 ml are sown on 25 g of soil. When seeding suspensions of fungi in selected dilution, in concentration corresponding to this dilution, the number of mycelial culture colonies sufficient to mycorrhizal formation grows on the soil: from the 5th dilution of T. Equestre with a concentration of 7.8±0.3·107 FR/cm3, 59 colonies are grown, from the 5th dilution of L. Rufus with a concentration of 8.1±0.2·107 FR/cm3, 60 colonies are grown, from the 8th dilution of S. Bovinus with a concentration of 10.2±0.1·109 FR/cm3, 76 colonies are grown.

EFFECT: invention makes it possible to increase the fragments of mycelium.
