COLUMN-BASED METHODS FOR CLEANING VECTOR BASED ON AAV Russian patent published in 2022 - IPC C12N15/864 

FIELD: biotechnology.

SUBSTANCE: in the present document, methods for cleaning vector particles based on a recombinant adeno associated virus (hereinafter – rAAV) are described. Methods include collection of cells, their lysis, lysate treatment with nuclease, at least 2 stages of column chromatography, and filtration of the resulting eluate. Column chromatography stages include cation-exchange chromatography, anion-exchange chromatography, exclusive chromatography and/or affine chromatography of AAV separately or in a combination and in any order. Proposed methods can be scaled to larger volumes and applied to a wide range of serotypes/capsid AAV options.

EFFECT: methods provide for the reduction in the formation of impurities during rAAV obtaining.
