FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology and agriculture and consists in preliminary incrustation of legume seeds using biopolymer shells based on strains of beneficial microorganisms and polysaccharides of biological and microbial origin. Method for incrustation of legume seeds at the first stage involves separately cultivating strains of rhizobia Bradyrhizobium japonicum, Rhizobium leguminosarum bv. viciae, or Mesorhizobium cicer, and cultivating a producer strain of extracellular polysaccharides Paenibacillus mucilaginosus to accumulate bacterial exopolysaccharides (EPS); at the second stage, stabilising the rhizobia with a solution of trehalose, wherefor a sterile solution of trehalose is added to the liquid culture of Bradyrhizobium japonicum, Rhizobium leguminosarum bv. viciae, or Mesorhizobium ciceri, the resulting suspension is held for 1 to 3 hours at 20 to 25°C, followed by cooling at 4°C; at the third stage, sterilising the culture of the producer strain of extracellular polysaccharides of the Paenibacillus mucilaginosus class by autoclaving; at the fourth stage, preparing a liquid biopolymer stabiliser, wherefor at least 60% of inactivated culture fluid of Paenibacillus mucilaginosus is added to the sterile 0.85% solution of sodium chloride and the components are mixed; at the fifth stage, producing a stable form of the biological incrustator, wherefor the suspension of the stable culture of rhizobia, produced at the second stage, is mixed with the biopolymer stabiliser produced at the fourth stage and a 10% polyvinyl alcohol solution in the ratio 1:1:1, the mixture is held for 1 hour at 20 to 25°C; at the sixth stage, mixing the legumes seeds with a liquid biological incrustator in an amount of 2 to 5 wt.% of the seeds, followed by covering the treated seeds with an anticaking agent in an amount of 2 to 5 wt.% of the seeds; and at the seventh stage, subjecting the incrustated seeds to soft drying while constantly stirring at 40 to 42°C for 120 to 180 minutes to a moisture content of 12 to 15%.
EFFECT: proposed method for incrustation of legume seeds ensures the preservation of beneficial bacteria in the shell applied to the seed for at least 9 months in titers>104CFU/seed sufficient for the effective inoculation thereof during seeding, acting as an effective means of increasing the productivity of legumes and eliminating the need for liquid inoculants and other biological agents when seeding legumes.
