FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, in particular to genetic engineering, and relates to a new strain of bacteria, which is used to obtain recombinant interferon gamma (hereinafter INF-gamma) used in the medical industry. The recombinant plasmid pET32a-IFG144 - 5806 base pairs (bp) in size provides the synthesis of a protein having the amino acid sequence of SEQ ID NO: 1, 16.9 kDa in size and contains structural elements: the T7 promoter and the lacO operator, the synthetic nucleotide sequence of SEQ ID NO: 2 encoding IFN-gamma protein, ampicillin resistance gene (AmpR) and bacterial ampicillin resistance gene promoter (AmpR promoter) for selection of recombinant cells, bacteriophage replication origin fl (fl ori), lactose repressor gene LacI and bacterial promoter of the lactose repressor gene (LacIpromoter).
EFFECT: invention makes it possible to increase the efficiency of obtaining gamma interferon protein, namely, to increase the protein yield from the biomass of a strain with a purity of the final product of at least 97% by creating a recombinant strain of Escherichia coli - an IFN-gamma producer, obtained on the basis of a constructed recombinant plasmid, which provides an increased yield of IFN -gamma by increasing the amount of biomass obtained during cultivation by 6-10 times.
