METHOD OF TESTING MEDICINAL SUBSTANCES FOR BELONGING TO SUBSTRATES OF POLYPEPTIDE TRANSPORTING ORGANIC ANIONS 1B1 Russian patent published in 2024 - IPC G01N33/50

Abstract RU 2815047 C1

FIELD: pharmacology.

SUBSTANCE: disclosed is a method of testing medicinal substances for belonging to substrates of a polypeptide transporting organic anions 1B1, involving the use of a human embryonic kidney cell line transformed with type 5 adenovirus, version 293, and a human embryonic kidney cell line transformed with type 5 adenovirus, version 293, which have been further transfected with a gene encoding a polypeptide transporting organic anions 1B1 with an attached green fluorescent protein, incubating cells with a transport medium containing a test substance in a final concentration of 1 mcM, for 5 minutes and determining the concentrations of this substance in cell lysates, wherein if concentration of test substance in lysate of human embryonic kidney cells transformed by type 5 adenovirus, version 293, which were additionally transfected with a gene coding a polypeptide transporting organic anions 1B1 with attached green fluorescent protein higher than its concentration in human embryonic kidney cell lysate transformed with adenovirus type 5, version 293, 2 and more times test substance is a substrate of a polypeptide which transports organic anions 1B1.

EFFECT: invention provides an accessible and effective method for assessing the belonging of medicinal substances to substrates of a polypeptide transporting organic anions 1B1.
