BIOLOGICALLY ACTIVATED DRUGS BASED ON CYTOKINES AND METHODS OF USING THEM Russian patent published in 2024 - IPC C07K14/54 C07K14/55 C07K2/00 A61K38/00

Abstract RU 2819307 C2

FIELD: biotechnology.

SUBSTANCE: present invention relates to novel interleukin-based peptide structures, and can be used in medicine. According to the present invention, a cytokine-based biologically activated drug construct (VitoKine) platform is provided, aimed at reducing systemic toxicity and providing a wider therapeutic use of interleukin-15 (IL-15) for the treatment of cancer, autoimmune diseases, inflammatory diseases, viral infection, transplantation and various other disorders. New VitoKine constructions according to the present invention contain: (1) a D1 domain of a tissue or disease focus group; (2) a D2 domain of a biologically activated group; and (3) masking group domain D3.

EFFECT: it is important that the VitoKine structure "active grouping" inertness is maintained until its local activation by proteases, the regulation of which is increased in the tissues affected by the disease, will limit the binding of the active group to its receptors or targets on the periphery or on the surface of cells and tissues not affected by the disease, preventing excessive activation of its pathway and reducing undesirable toxicity caused by binding to a target outside the target tissue; in addition, the inertness of the active group of VitoKine before activation by proteases will significantly reduce the possible clearance caused by binding to the antigen or target, and thus, increase the half-life in vivo and improve biodistribution, bioavailability and therapeutic efficacy.
