FIELD: biotechnology.
SUBSTANCE: disclosed is a method of producing a heterogeneous biocatalyst based on cellulase immobilized on a cation-exchange resin Purolite C100 in H-form, involving adsorption immobilisation of cellulase recovered from Aspergillus niger. Before immobilisation, cation-exchange resin Purolite C100 in H-form is held for 12 hours at room temperature in 0.1M acetate buffer with pH=4.5 based on 1 g of cation-exchange resin using 20 ml of buffer, resin is separated from buffer by decantation, and then immobilization is carried out by adding 20 ml of cellulase solution in 0.1 M acetate buffer with pH=4.5 in a cellulase concentration of 10 mg/ml to 1 g of cation-exchange resin maintained in buffer and incubation is carried out with stirring at 250 rpm for 1.5 h, then the external solution is separated from the cellulase immobilized on the cation-exchange resin by centrifugation at 3000 rpm for 5 minutes and subsequent decantation of the external solution; formed biocatalyst is washed by dialysis for 8 hours against 0.2 M phosphate-citrate buffer with pH 6.5, based on 1 g of biocatalyst, 400 ml of 0.2 M phosphate-citrate buffer is used, after which the buffer is replaced with a portion of buffer in volume of 400 ml and dialysis is continued for another 16 hours until there is no free cellulase in the washing solution.
EFFECT: invention enables to obtain an immobilized heterogeneous water-insoluble biocatalyst, washed from the non-immobilized form, with high specific activity.
