FIELD: biochemistry, analytical chemistry. SUBSTANCE: milled cereal seeds were extracted with water at pH 7.6 and precipitate is separated. After ion-exchange chromatography on DEAE-sorbent enzyme is desalted with ammonium sulfate (saturation degree is 0.7), dialyzed and dried. Prepared carboxylesterase is used for making one- and two-elemental matrix consisting of the backing with applied components of substrate/indicator system. Matrix is universal agent for detection of organophosphate compounds (OPC) of the general formula in water, air and extracts with organic solvents. Except for, carboxylesterase prepared from the milled cereal culture grains can be used for quantitative determination of OPC. Assay involves the determination of enzyme activity with or without analyzed sample, in reaction of substrate hydrolysis of the general formula ROC(O)R1 and ROC(S)R1 where R - phenyl, 1- and 2-naphthyl, R1- CH3, C2H5.. Plant origin carboxylesterase can replace esterases of animal and microbial origin (butyrylcholine esterase, acetylcholine esterase, etc.) used for the simplest procedures for OPC assay.
