FIELD: biotechnology.

SUBSTANCE: disclosed is a method of producing carboxyl esterase, involving grinding plant material of cereal crops, extraction at pH=7.5 for 1 hour at 28–30°C, then the suspension is held for 18–20 hours, the supernatant is drained, the precipitate is squeezed, filtered, and the pH of the solution is adjusted to 7.6±0.05 at 24–26°C and stirring; cleaning is carried out by adding DEAE-sorbent in portions of 0.5–1.0 g with intense stirring, after which the suspension is stirred for 1 hour, then the sorbent is deposited for 1 hour, decanting, washing the precipitate three times with 0.05M phosphate buffer solution, then with an aqueous solution of sodium chloride; salted out with sodium chloride, centrifuged; matrix for detection of organophosphorus compounds of general formula in substrates, in form of esters of general formula ROC(O)R1 and ROC(S)R1, where R is phenyl, naphthyl 1 and 2, R1 is methyl or ethyl, made in the form of a substrate from treated with a chromium mixture, washed with distilled water and dried crushed neutral glass of grade HC-1 with a fraction size of 0.40–0.63 mm of a certain chemical composition, using the obtained carboxyl esterase; method of making matrix, including crushing and sieving of neutral glass of HC-1 grade with separation of fractions with size of 0.40–0.63 mm, treatment of separated fractions with a chromium mixture, washing with distilled water, drying, impregnation with carboxyl esterase enzyme in ratio: 3 parts of enzyme in ml and 10 parts of crushed glass in g and drying.

EFFECT: inventions provide increased activity of carboxyl esterase, broader functional capabilities of matrices with given enzyme activities.
