SOLID MEDICINAL FORM OF THE PROLONGED ACTION FOR CARDIOVASCULAR DISEASE TREATMENT Russian patent published in 1997 - IPC

FIELD: medicine, pharmacy. SUBSTANCE: solid medicinal form of the prolonged action releases an active substance continuously for 15 h independently of gastroenteric tract pH value. Composition has core containing metoprolol and shell and consists of a lot of globules (size is 0.25-2 mm) core of that have at least 95 wt.-% metoprolol as salt (solubility in water is 600 mg/ml at 25 C). Active substance is released at velocity 3-7 wt.-%/h. Polymeric shell is applied on each globule. Preferable globule size is 0.35-1.0 mm. Metoprolol salt can be benzoate or sorbate of metoprolol S-enantiomer. Globule content can contain metoprolol salt applied on the insoluble core made of silicon dioxide. Insoluble core made of silicon dioxide can show size 0.1-1.0 mm, preferably 0.15-0.3 mm, and to be coated with metoprolol salt. The formed globules (size is 0.35-1.0 mm) were coated with polymeric shell. Polymeric shell can be made of ethylcellulose or its mixture with hydroxypropylmethylcellulose. Medicinal form can be made as gelatin capsule filled with globules coated with polymeric shell, or as tablet made of the pressed globules with pharmaceutical additions.
