FIELD: biotechnology, medicine. SUBSTANCE: the parent raw (blood serum) is diluted preliminary with buffer solution, subjected for ion-exchange chromatography on DEAE column at pH = 6.2-6.4 and immunoaffinity chromatography on sorbent with immobilized antibodies to the human alpha-fetoprotein. The source of these antibodies - polyclonal monospecific antibodies to the human alpha-fetoprotein being purified preliminary. Human alpha-fetoprotein is eluted from immunoaffine column with 0.01 M solution of hydrochloric acid and the obtained fraction of the human alpha-fetoprotein is purified additionally by affinity chromatography on columns with sorbent based on antibodies to the normal human blood serum proteins and sorbent containing rabbit immunoglobulins to the human alpha-fetoprotein followed by gel-filtration, concentration and lyophilic drying the end product. EFFECT: high purity of preparation.
