METHOD OF PRODUCING TRANSGENIC TYPHOID FLY, MUSCA DOMESTICA Russian patent published in 1999 - IPC

Abstract RU 2137362 C1

FIELD: biotechnology, genetic engineering. SUBSTANCE: invention relates to the development of system of genetic engineering vectors from pair plasmids: pHerm[NeoR] consisting of base-vector pBS+, flank sites of mobile member Hermes and gene determining the resistance to an antibiotic neomycin NeoR under control of promoter of heat shock protein Hsp-70 of Drosophila melanogaster and plasmid pHl pHt containing the native transpose sequence of mobile member Hermes. Invention proposes a procedure for introduction of plasmid mixture pHerm[NeoR] and pHlpHt in preblastodermal embryos Musca domestica. System is designed for introduction of foreign genes inserted into pHerm[NeoR], into Musca domestica embryos to obtain transgenic strains of typhoid flies. Fly larvae with integrated gene structure into genome acquires the resistance to synthetic antibiotic geneticin (G418). Culturing larvae on medium with geneticin (concentrations are 200-1 000 mcg/ml) provides the viability transgenic species only. Invention ensures to obtain transgenic fly-producers of valuable biologically active substances and control of natural insect populations. EFFECT: improved method of producing. 1 dwg, 4 tbl, 5 ex
