SUBSTANCE: composition has at least one active component, 0.5-60 weight% of at least one vehicle and 5-50 weight% of at least one water-soluble at physiological values pH alkalinizing agent. Vehicle is taken among inert matrices, hydrophilic matrices, their mixtures and lipid matrices with exception for mixtures containing polyacrylic acid and at least one hydrophilic matrix of cellulose type. Alkalinizing agent is taken among alkaline or alkaline-earth metal hydroxides, carbonates, hydrocarbonates and phosphates, sodium borate and organic acid basic salts. The second variant of composition is made as two layers being at least one layer has an active substance and vehicles and provides immediate release of this active substance after its administration. At least one the second layer has composition described above and provides the sustained-release of the same or the second active substance. By the first variant method of the composition preparing involves preparing the homogeneous mixture of indicated components, granulation of mixture and its tableting. By the second variant method of the composition preparing involves separate preparing homogeneous mixtures of layer components, their following tableting in device for multilayer tableting. Invention provide to develop medicinal forms with sustained-release of active substances in the case of administration of substances by small daily doses, preferably as a single daily dose. Therapeutic effect of substances is evident for prolonged time, for 12 h, not less. Invention can be used for preparing preparations for oral administration.
