FIELD: medicinal industry, preparative biochemistry, hormones. SUBSTANCE: invention relates to preparing highly purified monocomponent insulin of different origin. Crude insulin is subjected for two procedures of column reverse-phase chromatography: at pH 1.5-4.5 and at pH 6.5-9.5. Purification is carried out on nonpolar sorbents exhibiting mean pores size 100-500 and particles size from 10 to 100 mcm. Aqueous-organic solvents containing ionic modifying agents are used as eluents. Process of chromatography treatment is carried out at temperature 5-30 C. Method provides preparing insulin of high quality containing 98.5% of basic substance, not less, 0.5% of desamido-A21-insulin, not above, 1.0% of unidentified impurities, not above, 1.0 mln-1 of proinsulin, not above.
