FIELD: virology, molecular biology. SUBSTANCE: noninfectious mutated virus Sindbis (Sindbis E1-SER62) where conservative amino acid cysteine in position 62 of protein E1 is substituted for serine is obtained. This mutation is located in viral protein domain determining thiol reductase/protein disulfide isomerase activity. Fragment of recombinant DNA that presents sequence of mutated noninfectious virus Sindbis is fused into plasmid that is amplified in E. coli and the obtained RNA is used for transfection of mammalian cells which produce the above described protein.
