FIELD: organic chemistry, nucleosides, chemical technology. SUBSTANCE: invention relates to method of synthesis of 5'-deoxy-5-fluorouridine of the formula (I) that involves the following steps: (a) interaction of 2', 3'-O-isopropylidene-5-fluorouridine of the formula (II) with functional derivative of sulfonic acid of the formula R-SO3H where R means (C1-C4)-alkyl, trifluoromethyl, unsubstituted, mono-, di- and tri-substituted phenyl group; (b) interaction of derivative of the formula (IV) with alkaline or alkaline-earth metal iodide; (c) hydrolysis of derivative of the formula (V) in acid medium and (d) reduction of derivative of the formula (VI) with hydrogen or hydrogen donor. Invention uses chloride, anhydride or mixed anhydride as the source of functional derivative of sulfonic acid. Reduction reaction is carried out by catalytic hydrogenation.
