METHOD FOR DETECTING ACTIVITY OF ALKALINE PHOSPHATASE IN BLOOD SMEARS OF CATTLE AND POULTRY Russian patent published in 2003 - IPC

FIELD: veterinary medicine. SUBSTANCE: the present method deals with preparing blood smears, their fixation and treatment with buffer-incubation mixture, incubation of blood smears followed by drying and carrying out microscopy. Moreover, to prepare buffer mixture one should apply 0.025 M sodium tetraborate supplemented with 4.6-5 ml 0.1 M hydrochloric acid to provide pH being 9.18-9.19. Incubation of blood smears should be conducted in darkness for 30 min at 18-20 C. Then one should wash out blood smears with running water for 10 min, rinse with distilled water and dry. Cellular nuclei of neutrophilic granulocytes should be stained to achieve red color with a dye - 0.5%-safronin aqueous solution or neutral red for 0.5-1 min. According to the quantity of brown-stained granules in neutrophilic cytoplasm one should detect activity of alkaline phosphatase enzyme by average cytochemical index. The present innovation is more simplified, accurate, enables to increase the level of immunological trials in cattle and poultry. EFFECT: higher efficiency. 1 cl
