CYTOTOXIC COMPOSITION FOR DAMAGE OF TARGET-CELLS Russian patent published in 2003 - IPC

Abstract RU 2218938 C1

FIELD: cellular biology, biophysics. SUBSTANCE: invention proposes cytotoxic composition for damage of target- cells consisting of component for recognition of target-cells and realization by them receptor-mediated endocytosis, component for directed intracellular transport into nucleus, component-carrier for assembling and stabilization of composition and properly damaging agent astatine-211 bound with protein moiety of composition in the ratio providing the cytotoxic effect. Prepared specific species of the composition characterizing by structure (modified sequence of signal of nucleus localization of T-antigen SV-40 component for intracellular transport) - (β--galactosidase-carrier) (insulin component for recognition and endocytosis) (astatine-211) and by ratio astatine-211/protein that is sufficient for preparing the composition with specific radioactivity 5 MBq/mg, not less, allows owing to the directed effect to obtain the expressed cytotoxic effect (in part, in cells of liver hepatoma) in using the preparation with specific activity by about 10 times lesser as in the case of free radionuclide. Invention provides possibility for damage of cells of definite type, for example, tumor cells in relatively low concentrations of radionuclide and minimal effect on surrounding tissues. Invention can be used in medicinal practice and pharmacology. EFFECT: enhanced effectiveness, valuable properties of composition. 6 cl, 3 dwg
