METHOD FOR PREPARING IMMUNOGLOBULIN PREPARATION Russian patent published in 2005 - IPC

SUBSTANCE: method involves extraction of alcoholic precipitate B (Cohn's fraction 111) with distilled water, purifying extract from lipoproteins with chloroform and sodium chloride up to the final concentration 0.9%, centrifugation, precipitation of immunoglobulins with alcohols, repeated centrifugation followed by addition of a stabilizing agent to a precipitate, carrying out clearing and sterilizing filtration of the end product and its lyophilic drying. Extraction of the alcoholic precipitate B is carried out in 12-fold volume of water and after addition of sodium chloride to the final concentration 0.9% pH value is brought about to from 4.0 to 5.0, then chloroform is added up to the final concentration 3.0% and the mixture is kept at 0-3°C for 14 h. After centrifugation a precipitate is discarded and clearing filtration is carried out. To precipitate immunoglobulins 50% solution of polyethylene glycol is added to the final concentration 12% or 26% ethyl alcohol. After centrifugation a precipitate is dissolved in 0.3 M glycine, pH value is brought about to 4.6 and after clearing and sterilizing filtration virus is inactivated at room temperature for 20-25 days. Method provides enhancing quality in purifying immunoglobulins from impurities, enhanced yield of the end preparation, providing high solubility, stability and safety of antibodies of all classes in the end preparation. Invention can be used in medicine for treatment and prophylaxis of severe infectious diseases and immunodeficiency states.
