FIELD: medicine, chemical-pharmaceutical industry, pharmacy.
SUBSTANCE: invention relates to a method for isolating biologically active substances from animal organs. Method involves freezing brain cortex from slaughtering animals, homogenization of frozen tissue and extraction of homogenate with acetic acid solution in the presence of zinc chloride followed by separation of homogenate, treatment of supernatant liquid with acetone, washing the formed precipitate with acetone and drying the precipitate. Then polypeptides from dried powder are extracted with water for injection followed by filtration, sterilization and lyophilization of the end product. However, in contrast to the known methods parts of procedures are varied. For producing the complex method involves using cattle brain cortex before 12-month age. The procedure for freezing the brain tissue is carried out at temperature -18°C, not above. The frozen brain cortex tissue is used not early in 2 months and not later in 8 months. Extraction of polypeptides from dried powder is carried out with water for injection for two stages. At the first stage extraction is carried out at pH 5.5-6.6 and temperature 18-22°C. After separation of extract method involves carrying out the second extraction stage of polypeptides with water for injection under the same conditions. Proposed method provides enhancing quality and the percent yield of the end product. Pharmaceutical agent used for normalization of brain functions comprises the end product as an active component prepared by proposed method and representing the complex of biologically active polypeptides of molecular mass 500-15000 Da, isoelectric point value at 3.5-9.5 and the absorption maximum in UV-spectrum at wavelength 275 ± 6 nm. The proposed end product can be used in medicine, pharmacy and veterinary science as a pharmaceutical agent able to normalize functions of the brain.
