FIELD: medicine.
SUBSTANCE: the innovation deals with immunoluminescent studying of smears-preparations out of the samples under testing dyed with diagnostic fluorescent immunoglobulin against B.cepacia, interacting with B.cepacia and not reacting for the presence of other types of burkholderia and pseudomonas. The above-mentioned immunoglobulins are obtained based upon gamma-globulin fraction isolated out of rabbit's hyperimmune serum due to precipitation of ammonia sulfate and fluorochromium-labelled. Hyperimmune serum should be isolated out of rabbits' blood which, also, should be immunized with the help of the mixture of B.cepacia and Freund's adjuvant. Antigens are obtained out of the cells of B.cepacia strain 25416 ATCC which should be grown on solid nutritive value to be then washed with 0.9%-NaCl solution at pH being 7.2, sterilized to be cooled up to 40°C with acetone and dried under the pull. Then dry cells of B.cepaci should be suspended in 0.9%-NaCl solution to be then treated with ultrasound. Disintegrant should be applied for immunization in rabbits. Activity of sera should be evaluated in reaction of immunodiffusion in gel with antigenic complex. At the titer of serum being 1:32 one should conduct bleeding in animals. The innovation provides the simplicity and efficiency of differentiation, for about 2-4 h it is useful to provide simplicity and efficiency of tested and provides the content of B.cepaia.
