FIELD: analytical chemistry.

SUBSTANCE: invention relates to a method for determination of endotoxin level in solution of colored proteins, in particular, to a method for quantitative determination of endotoxin in 10% hemoglobin solution. Method for determination of endotoxin level in hemoglobin solution involves using the kinetic turbidimetric LAL (Limulus amoebocyte lysate), assay at wavelength 650 nm or 660 nm using optical filter. Method involves the following steps for modifying a sample: (a) dilution of 10% weight/volume hemoglobin solution to the concentration 10 mg/ml; (b) incubation of the prepared diluted solution at the temperature range 60-80°C for 10-20 min; (c) dilution of the incubated solution with LAL-water up to the concentration 2.5 mg/ml; (d) testing the prepared solution using kinetic turbidimetric LAL analysis at wavelength 650 nm or 660 nm using optical filter; (e) recording prepared concentrations of endotoxin using standard plot prepared for turbidimetric LAL analysis. Invention provides the development of a method for determination of endotoxic level in solution of colored proteins that overcomes shortcomings of the known methods since proteins and among of them hemoglobin interferes mainly with test-system and shows false-positive or false-negative results.
