METHOD OF PRODUCING MONOSPECIFIC ANTIBODIES TO PLANT CELL PROTEINS Russian patent published in 2011 - IPC C07K16/16 C12P21/08

Abstract RU 2410395 C2

FIELD: chemistry; biochemistry.

SUBSTANCE: invention relates to biotechnology and is meant for producing monospecific antibodies to plant cell proteins. Purified protein antigens are produced via chromatography and electrophoresis in polyacrylamide gel. The antigen used is a marker protein of meristematic cell of wheat. Before immunisation, the obtained protein antigens are incubated in tris-HCl with a buffer solution containing sodium dodecylsulphate and 2-mercaptoethanol for 12-24 hours at 2-10°C. Laboratory animals are immunised three times with two-week breaks. The first immunisation is done in popliteal lymph nodes with a portion of the crushed gel containing the antigen with Freund's complete adjuvant, and the second and third immunisations are done in peritoneum active sites with a portion of the crushed gel with incomplete Freund's adjuvant. Monospecific serum, which contains the antibody to the investigated antigens, is obtained from the blood of the immunised animals. The antibodies are purified by fractionating antiserum with caprylic acid followed by desalination and cation-exchange chromatography.

EFFECT: invention increases specificity and titre of obtained antibodies owing to additional purification operations.
