FIELD: medicine.
SUBSTANCE: claimed invention relates to a method of obtaining and purification of orthopoxvirus. The claimed method includes the following stages: preparation of a culture of packaging cells; infecting the culture of packaging cells with orthopoxvirus; cultivation of the infected packaging cells until the orthopoxvirus progeny is obtained; incubation in the presence of one or more nucleases until nucleic acids are destroyed; separation of orthopoxviruses from the supernatant of the culture and/or packaging cells; addition of monovalent salts in the concentration from 200 to 300 mM under conditions, suitable for the inhibition of nuclease activity and for the prevention of adsorption of the said orthopoxviruses to an anionic adsorbent; realisation of a contact of the obtained mixture with anion-exchanging adsorbent under conditions, suitable for trapping nucleic acids. The characterised invention can be used for the elaboration of vaccines.
EFFECT: obtaining and purification of orthopoxvirus.
