METHODS OF IMPROVING PROTEIN EFFICIENCY Russian patent published in 2015 - IPC C12P21/02 C12N15/10

Abstract RU 2569106 C2

FIELD: chemistry.

SUBSTANCE: invention relates to the field of biotechnology and represents a method of obtaining at least one version of a protease with an improved washing efficiency in a powder-like or liquid detergent composition, which has pH between 5 and 12.0, in comparison with an initial protease. The said method includes: a) determination of optimal total charges of the protease surface for the washing activity in a powder-like or liquid detergent composition, which has pH between 5 and 12.0, by testing in the said medium of multiple versions of substituted proteases, which have a change of the total charge of the molecule surface, with the optimal total charges of the protease surface representing charges -2, or -1, or +1, or +2 relative to the initial protease; b) modification of at least one amino acid residue in one or several positions, which have a solvent-accessible surface larger than 50%, on the surface of the said initial protease to obtain at least one version of the protease, which has the more positive or more negative surface charge in comparison with the initial protease for the protease versions to have the optimal total surface charge as determined at stage a); and c) estimation of the washing efficiency of at least one version of the protein from stage b) to identify the version of the protein demonstrating the improved washing efficiency in the said test of interest in comparison with an initial protein.

EFFECT: invention makes it possible to obtain proteases with the improved washing efficiency in the powder-like or liquid detergent composition, which has pH between 5 and 12,0 in comparison with the initial protease.
