FIELD: biotechnology.

SUBSTANCE: method comprises inoculation of the test material in a container with a two-phase nutrient medium containing a liquid and solid phase in a volume ratio of 1:3. The test material is added into the liquid phase of the two-phase nutrient medium and the culture is incubated three times. The first time - for 4.0 hours. Then the contents of the liquid phase are stirred. The container with a two-phase nutrient medium is placed in an inclined position so that the liquid phase of the nutrient medium wets fully the entire surface of the solid phase, and the culture in the inclined position is incubated again for 0.5 hours. Then the container with the culture is placed in a vertical position and incubated for the third time for 12.5 hours.

EFFECT: invention enables to reduce the time of cultivation and to increase the number of colonies of diphtheria bacteria with their low content in the test material.
