FIELD: chemistry, biochemistry.

SUBSTANCE: invention refers to medical microbiology, specifically to laboratory diagnostics of infection agents. There is declared method of differentiating strains C.diphtheriae concerning biovar gravis, based on revealing amylase (amy) gene fragment sized 839 base pairs with using polymerase chain reaction with primer pair AmyFl 5'-TTGATCCGCACCTCGGTTCC-3' and AmyRl 5'- GTACCTTGTCATCATTATCA-3' specific to this fragment. It is followed with electrophoresis of amplification and differentiation products by comparing electrophoretic mobility of these amplicons to that of marker DNA fragments. If specific flashing strip at level 839 base pairs is observed, the given strains are registered as C.diphtheriae biovar gravis.

EFFECT: provided accurate, accelerated and high-avid method of differentiating strains Cdiphtheriae by genetic determinant, responsible for enzyme synthesis of amylase for the purpose of revealing strains belonging to biovar gravis, being a necessary component of epidemiological control of diphtheritic infection, thus ensuring timely and adequate treatment.
